If we were to add many additional custom profile fields for members, would these fields be searchable via the advanced search?

1. Yes, is you make those fields 'searchable' when you create them.

2. Yes, using the CSV/text importer. You can import username, password and email address.

Nope, just those unless you want to modify the script yourself.

Probably not but it depends on what exactly you want to import and how experienced you are with PHP/MySQL programming.
